Effect of NF-κB and CD40 blockade on the IL-17-mediated induction of (a) IL-6 and (b) IL-8 in FLS from two patients with RA. FLS were cultured in triplicate with or without 10 ng/ml IL-17 for 24 hours and assayed for the production of IL-6 and IL-8 by sandwich ELISA. Effects of NF-κB blockade and CD40 ligation were investigated by adding 3 μM PDTC and 10 ng/ml sCD40L, respectively, in IL-17-stimulated culture.
