Effect of MAPK blockade on the IL-17-mediated induction of (a) IL-6 and (b) IL-8 in FLS from two patients with RA. FLS were cultured in triplicate with or without 10 ng/ml IL-17 for 24 hours and assayed for the production of IL-6 and IL-8 by sandwich ELISA. Effects of blocking MAPK activation were investigated by adding 1 or 10 nM SB203580 at the time of IL-17 stimulation.
