Apoptosis of acinar cells from NOD submandibular glands. (a) Chromatin condensation and mRNA and protein expression of (b) Bax and (c) TP53INP1 were determined in acinar cells from non obese diabetic (NOD) at 16 and 8 weeks of age and 16 week old BALB/c mice. (c) The light grey bar corresponds to TP53INP1β while TP53INP1α is in dark bars. The count of condensed nuclei at 400× after Hoechst staining and RT-PCR or immunoblotting for apoptotic mediator expression were performed as indicated in Material and methods. These images are representative of similar experiments run with independent NOD and BALB/c acinar samples. Results shown on the right side of representative photographs represent the percentage of apoptotic cells as mean ± standard error of the mean (SEM) of at least six experiments. (a) *P < 0.05 vs. BALB/c 16 weeks. The intensity of mRNA or protein bands relative to GAPDH or actin, respectively, were depicted as the mean ± SEM of at least three experiments.
