Human glucose-6-phosphate isomerase-specific Th17 cells use TCRVβ8.1 8.2. Many glucose-6-phosphate isomerase hGPI325--339-specific Th17 cells use TCRVβ8.1 8.2. Mice were immunized with 10 μg hGPI325--339, and draining lymph node cells on day 6 were stimulated with 20 μM hGPI325--339 in vitro. (a) Ratios of TCRVβ repertoire to CD4+ T cells. The TCRVβ repertoire of CD4+ T cells was analyzed by flow cytometry: before stimulation with hGPI325--339 in vitro for 72 hours, and after stimulation. (b) GoldiStop was added in the last 4 hours of the 24-hour culture. Flow cytometry analysis for IL-17 was gated in CD4high cells. Representative data of two independent experiments.
