  1. Groups of three DBA/1 mice were administered intra-articularly either adenoX-rCB11-A9 or adenoX-LacZ and the mice were immunized with type II collagen/complete Freund's adjuvant (CII/CFA) three days after the injection. Draining lymph node cells were harvested 14 days after the immunization and cultured (5 × 106 cells/ml) with 100 μg/ml of the indicated antigens, either Ova (negative control), the mouse collagen immunodominant wild type peptide, or Purified Protein Derivative (PPD) (positive control). Supernatants were collected 72 hours later and analyzed for the presence of the indicated cytokines. Values are expressed as picograms per ml and represent the mean values for each group.
  2. Cytokines IL-2, IFN-γ, IL-17, IL-5, and IL-10 were all significantly higher in response to the mouse collagen immunodominant peptide in the adenoX-lacZ treated mice compared with mice treated with adenoX-rCB11-A9 (P ≤ 0.05). On the other hand, IL-4 was significantly greater in the mice treated with aden-rCB11-A9 (P ≤ 0.05).
