(b) Treg isolated from a systemic lupus erythematosus (SLE) patient as CD4+ CD127-, then sorted as CD25+ or CD25- after stimulation and expansion with transforming growth factor beta (TGFβ) and retinoic acid (RA). The CD25+-sorted population was approximately 80% FoxP3+ CD25+, while the CD25--sorted population was more than 80% FoxP3+ CD25-. (c) Proliferation of CD25+ and CD25- Treg isolated from a same patient in the presence of standardized peripheral blood mononuclear cells (PBMCs) at the same dilution (1:4), in the presence of anti-CD3 and anti-CD28 for six days, showing a robust response of the CD25+ as opposed to the CD25- Tregs.
