PI3K is involved in CCN4-mediated IL-6 production in synovial fibroblasts. (A-C) OASFs were pretreated for 30 minutes with LY294002 (10 µM) and Wortmannin (5µM) followed by stimulation with CCN4 for 24 hours. Media, total RNA, and protein were collected, and the expression of IL-6 was analyzed by qPCR, ELISA, and Western blot. (D, E) OASFs were transfected for 24 hours with PI3K mutant followed by stimulation with CCN4 for 24 hours. Media and total RNA were collected, and the expression of IL-6 was analyzed by qPCR and ELISA. (F) Cells were incubated with CCN4 for the indicated time intervals, and PI3K phosphorylation was examined by Western blot. Results are expressed as the mean ± SEM (n = 3). *, P <0.05 compared with control; #, P <0.05 compared with the CCN4-treated group.
