The effect of lysophosphatidic acid receptor 1 on the expression of adhesion molecules on fibroblast-like synoviocytes. Rheumatoid arthritis (RA) fibroblast-like synoviocytes (FLSs) were pretreated with a lysophosphatidic acid (LPA) receptor 1 inhibitor (LA-01; 0, 1 or 10 nM) for 30 minutes, then the cells were stimulated with 10 ?M LPA for 12 hours. Cells were stained with allophycocyanin-conjugated monoclonal antibody (mAb) against vascular cell adhesion molecule (anti-VCAM) or phycoerythrin-conjugated mAb against intercellular adhesion molecule (anti-ICAM). Allophycocyanin- or phycoerythrin-conjugated mouse immunoglobulin G1 (IgG1) was used as a control. The expression of VCAM and ICAM on FLSs was analyzed by flow cytometry. Filled histogram (gray): isotype control; open histogram (black line): VCAM or ICAM.
