Pam3Cys- and poly:IC-induced production of IL-1β, IL-6, TNFα in peripheral blood mononuclear cells (PBMCs) sampled during active disease and IL-1-blocking treatment. PBMCs of patients with Schnitzler’s syndrome (SchS) without NLRP3 mosaicism were sampled during anakinra treatment, a symptomatic episode, at several time points during a canakinumab trial, and during relapse after canakinumab withdrawal. These PBMCs and those of healthy controls were stimulated with Pam3Cys 10 μg/ml (a), or poly:IC 5 μg/ml (b) for 24 hours, and supernatants were collected for ELISAs of IL-1β, IL-6 and TNFα concentrations.
