Calcium dependency of PAD reduced by glutathione. Microtitre plates were coated with human fibrinogen and incubated for 3 h at RT with rhPAD2 (300 ng/ml) or rhPAD4 (3000 ng/ml) and either 1 mM dithiothreitol (DTT) (open symbols) or 10 mM reduced glutathione (GSH) (closed symbols) in the presence of CaCl2 at concentrations ranging from 20 μM to 10 mM. Citrullination of fibrinogen was measured using mAb 20B2 as the detecting antibody.
