Effect of prophylactic URB597 treatment on the development of secondary allodynia after MIA injection. Intra-articular injection of MIA significantly reduced von Frey hair withdrawal threshold on day 1 (a) and day 14 (b) after injection compared with saline-injected knees. Treatment with URB597 (0.3 mg/kg) had no effect on day 1 secondary allodynia while prophylactic URB597 abolished chronic MIA-induced tactile hypersensitivity on day 14 compared with animals treated with a similar regimen of vehicle. **P < 0.01, *P < 0.05 unpaired two-tailed t test; n = 8–14.
