Capacity of Dow2 to suppress retinal antigen-specific T cell activation in vitro. IFN-γ production was measured by ELISA in culture supernatants of (a) splenocytes isolated from Dow2- or rat IgG-treated experimental autoimmune uveitis (EAU) mice (day 21 after immunization; n = 3 mice per group), cultured with or without Interphotoreceptor retinoid-binding protein (IRBP) retinal peptides for 48 h; and (b) intraocular cells from EAU mice with ocular inflammation (n = 12) cultured with IRBP retinal antigens and Dow2 or 17A2 for 48 h. Results of triplicates are presented as the mean ± SEM. *P < 0.05, **P < 0.005, ***P < 0.0005
