HCQ treatment inhibits ionomycin-dependent intracellular calcium mobilization. Jurkat cells (left) and purified CD4+ T cells (right) were pretreated with various concentrations of HCQ (1 × = 600 ng/ml) for 24 hours, loaded with a calcium-sensitive dye, and then activated by 1 μg/ml ionomycin (as indicated by the arrow). Before and after stimulation, changes of intracellular free Ca2+ concentration (delta fluorescence) versus time were monitored by spectrofluorometry at different time points. These results represent one assessment; more than six independent experiments had similar results. HCQ hydroxychloroquine
