Reproduced functional assays for synergistic cytokine induction. In place of siA20-1, two other anti-TNFAIP3 siRNAs with different target sequences, siA20-2 and siA20-3, were used in HeLa cells to demonstrate the absence of off-target effects on synergistic induction of NF-κB-mediated cytokine expressions. After treatment with TNFα for varying periods, CCL2, CXCL8, and IL6 mRNAs were measured against GAPDH mRNA using qPCR. See the legend of Fig. 4
