SF activation is independent of IFNγ and TBX21. a, b CCR6+ subpopulations were cultured and analyzed as in Fig. 4, but with 10 μg/ml neutralizing IFNγ antibody or isotype control. c Total CCR6+ Th memory cells were transduced with a scrambled shRNA (sh-scr) or two different TBX21 shRNA (sh1-TBX21 and sh2-TBX21). Stably transduced CCR6+ Th memory cells were cultured for 3 days with SF under stimulation of anti-CD3 and anti-CD28. Expression of TBX21 and EOMES was measured using RT-PCR on day 3. d Cytokine expression in culture supernatant from c was determined after 3 days using ELISA.
