Scheme for the synthesis of CPC-IGF-1 formulation and MALDI mass spectrometry confirming conjugation of CPC to IGF-1. A IGF-1 was first modified with maleimide (IGF-1-Maleimide) through reaction with a bifunctional NHS-PEG2-Maleimide linker via targeting the primary amines on IGF-1, thereby adding the maleimide to IGF-1 through stable amide bonds. The maleimide-modified IGF-1 (IGF-1-Maleimide) was then reacted with CPC +14N consisting of a cysteine residue to form CPC-IGF-1 conjugate through a stable thioether bond.
