Effect of MT-7117 on human fibroblast activation. Healthy or SSc dermal fibroblasts were grown to confluence and then starved in a medium containing 1% fetal calf serum for 24 h. Afterwards, cells were stimulated with TGF-β (10 ng/mL) in the presence of MT-7117 or αMSH for 24 hours. A ACTA2 and COL1A1 mRNA expression in healthy dermal fibroblasts was determined by real-time PCR and normalized by GAPDH expression during a dose-response treatment of both compounds. B ACTA2 and COL1A1 mRNA expression in healthy and SSc dermal fibroblasts were determined by real-time PCR and normalized by GAPDH expression using 1000 nmol/L of both compounds.
