Peripheral blood mononuclear cells were cultured with medium only (lane 1), anti-CD3 antibody (10 μg/ml; lane 2) or anti-CD3 antibody (10 μg/ml) plus LY294002 (20 μM; lane 3) for 10–120 min. Cell lysates were analyzed for Akt activation by western blot analysis of total and Ser473-phosphorylated Akt (P-Akt) using specific antibodies. Levels of phosphorylated Akt were compared at each time point, after normalization to Akt and β-actin in the same sample.
