Expression and induction of IL-17R and IL-17RB in IL-17-stimulated FLS from six RA patients. (a) IL-17 dose-dependent changes in the levels of IL-17R and IL-17RB mRNAs. Three independent RA FLS cell lines were stimulated with various amounts of recombinant IL-17 (0 to 20 ng/ml), and subsequent changes in the mRNA levels of IL-17R and IL-17RB were assessed by RT-PCR at 6 hours after the onset of in vitro culture. The relative intensity of each PCR band was normalized against that of GAPDH. Values are the fold increase from the unstimulated cell in each FLS line. (b) Time-dependent changes in the level of IL-17R and IL-17RB mRNAs. Three independent RA FLS cell lines were stimulated with recombinant IL-17, and subsequent changes in the mRNA level of IL-17R and IL-17RB were assessed by RT-PCR 0, 1, 3, 6, 9, and 24 hours after the start of in vitro culture. The relative intensity of each PCR band was normalized against that of GAPDH. Values are the fold increase from the 0 hour measurement in each FLS line.
