Prednisolone and dipyridamole combine to suppress acute inflammation in vivo. (a) Lewis rats were treated orally with compounds as indicated (mg/kg) for 2 hours before challenge with lipopolysaccharide (LPS). Serum was collected 90 minutes later and tumor necrosis factor (TNF)-α quantitated by ELISA. *P < 0.01 versus the LPS control; aP = 0.06, bP = 0.98, cP = 0.81, dP = 0.59 versus the combination. (b) Mice were sensitized with the chemical irritant 2,4-dinitrofluorobenzene (DNFB). Five days later animals were administered test agents by oral gavage at the indicated doses (mg/kg) and challenged on the ear with DNFB solution. Change in ear thickness was measured 24 hours after challenge (Δ ear thickness [mm]). *P < 0.05 versus the vehicle control; aP = 0.02, bP = 1.0, cP = 0.02, dP = 0.05 versus the combination. Dipyridamole was dosed at 150 mg/kg. Error bars are + standard error of the mean. Statistical comparison is by analysis of variance with Tukey. Dp, dipyridamole; Pd, prednisolone.
