Akt is involved in the potentiation of IL-6 production by CCN4. (A-C) OASFs were pretreated for 30 minutes with Akti (10 µM) followed by stimulation with CCN4 for 24 hours. Media, total RNA and protein were collected, and the expression of IL-6 was analyzed by qPCR, ELISA, and Western blot. (D, E) OASFs were transfected for 24 hours with Akt mutant followed by stimulation with CCN4 for 24 hours. Media and total RNA were collected, and the expression of IL-6 was analyzed by qPCR and ELISA. (F) Cells were incubated with CCN4 for the indicated time intervals, and Akt phosphorylation was examined by Western blot. Results are expressed as the mean ± SEM (n = 4). *, P <0.05 compared with control; #, P <0.05 compared with the CCN4-treated group.
