Lipolysaccharide (LPS)-induced production of IL-1β, IL-6, and TNFα in peripheral blood mononuclear cells (PBMCs) sampled during active disease and IL-1-blocking treatment. PBMCs of patients with Schnitzler’s syndrome (SchS) without NLRP3 mosaicism were sampled during anakinra treatment, a symptomatic episode, at several time points during a canakinumab trial, and during relapse after canakinumab withdrawal. These PBMCs and those of healthy controls were stimulated with 1 ng/ml LPS for 24 hours, and supernatants were collected for ELISA of IL-1β, IL-6 and TNFα concentrations.
